AOD-9604 (Anti-Obesity Drug 9604) is a modified fragment of the Human Growth Hormone (HGH) molecule. Specifically, it is the last 15 amino acids of the HGH chain (residues 176-191) with an added Tyrosine amino acid at the beginning to stabilize it.[1]

Full HGH does many things in the body: it builds muscle, grows organs, raises blood sugar, and burns fat. Scientists isolated just the tiny piece of the HGH molecule responsible for fat burning and created AOD-9604. The result is a peptide that stimulates lipolysis (the breakdown of fat) and inhibits lipogenesis (the creation of new fat) without causing the dangerous side effects of full HGH, such as insulin resistance or organ growth.[2]

AOD-9604 is highly sensitive to insulin. If insulin is present in your blood (from eating carbohydrates), the fat-burning mechanism of AOD-9604 is completely shut down. Therefore, it MUST be taken in a fasted state.

In a 12-week Phase 2b trial, patients on 1mg/day of AOD-9604 lost an average of 2.6 kg, compared to 0.8 kg in the placebo group. While statistically significant, the weight loss was considered too modest by pharmaceutical standards, and the company eventually halted development for obesity.[6]
  • Osteoarthritis: Interestingly, AOD-9604 is currently being investigated for the treatment of osteoarthritis. Animal models have shown that intra-articular injections of AOD-9604 promote cartilage repair and reduce joint inflammation, likely through pathways independent of its lipolytic effects.[7]
