NAD+ is too large to easily cross the cell membrane, which is why taking pure NAD+ orally is ineffective. Instead, the body relies on the "salvage pathway" to recycle and build NAD+.

  • In 2019, researchers at Washington University discovered a specific transporter protein called Slc12a8 located in the gut. This transporter is specifically designed to shuttle intact NMN directly into cells, proving that NMN does not need to be broken down into NR to be effective.[6]

  • A 2023 randomized, double-blind, placebo-controlled trial (Yi et al.) tested doses of 300mg, 600mg, and 900mg of NMN in healthy adults. It found that NMN safely and dose-dependently increased blood NAD+ levels, improved the 6-minute walking test distance, and reduced biological age (measured via blood age calculators).[7]
  • Another trial in postmenopausal women with prediabetes showed that 250mg of NMN daily for 10 weeks significantly improved muscle insulin sensitivity, mimicking the effects of significant weight loss.[8]
