Russian scientists attached a Proline-Glycine-Proline (Pro-Gly-Pro) sequence to the C-terminus of the peptide. This specific tripeptide sequence is highly resistant to enzymatic degradation by peptidases in the blood and brain. This modification extends the half-life of Semax to several hours and allows it to survive the journey across the nasal mucosa and the blood-brain barrier.[7]

In Russia, Semax 1% solution is a standard-of-care treatment for acute ischemic stroke.

Q: Will Semax make me fail a drug test? A: Yes, if you are a WADA-tested athlete. It is banned in-competition. However, for standard workplace drug tests (which look for amphetamines, THC, opiates, etc.), Semax will not trigger a false positive.

Q: Why does my Semax burn when I spray it? A: Standard Semax should not burn. However, N-Acetyl Semax Amidate (NASA) is often suspended in a slightly acidic solution to maintain stability, which can cause a brief, mild stinging sensation in the nasal cavity.

Q: Can I take Semax every day forever? A: It is not recommended. While it is very safe, constantly forcing the brain to upregulate BDNF may eventually lead to TrkB receptor downregulation (the receptor that BDNF binds to). Cycling (e.g., 5 days on, 2 days off) is best practice.
